Deciphering the epigenetic basis of progenitor differentiation. The expression of GFP transgenes (green) under the control of the yeast GAL4::UAS expression system shows unexpected variegation in Drosophila ovarian follicle cells. Remarkably, the clonal variegation pattern is caused by unstable epigenetic inheritance during follicle cell progenitor divisions, and the pattern, which shows many fewer small clones than large clones, reveals increasing epigenetic stability as progenitors approach their final divisions and start to differentiate. Dominant modifiers of GFP variegation were screened to identify genes affecting epigenetic maturation. Multiple new genes were identified that encode chromatin modifiers, cell cycle regulators and transcription factors. The nature of these modifiers supports previous evidence that the inability to form H3K4me3 modifications at key genes limits the ability of follicle progenitors to initiate differentiation independent of Notch signaling, while new genes affecting transcription and the cell cycle play previously unappreciated roles.
